We have tested 20x/1.0 objectives from Zeiss, Leica and Olympus on a new two-photon microscope (980 nm, no cover slip) and the results are - not great. All of these objectives have good IR transmission and are marketed for 2p, but I start to suspect they are still optimized for visible light. This is very disappointing. Objectives with the same NA and correction collar provide much better resolution (at a different price point). Have you used any of these objectives for high resolution 2p imaging? Send me you thoughts, prayers and PSFs, please.
